what are you trying to mod "hack" is a very general term are you looking for root. hardware mods,

Could I install a marketplace like AppBrain and if so would apps from the market work without a touchscreen? Can the device be rooted and/or custom ROMS installed and what capabilities would that enable beyond the stock eReader function. I don't see a need for an eReader, can it be turned into more of a general computing device? In short, can I turn it into an Android netbook that doesn't fold?

Doubtful. The Book is Linux, not android. I demoed one in the store and (IMHO) it is pretty flimsy.
